Cornus Compound Loganin Suppresses Expression of Fat Cell Differentiation Genes in Cell and Mouse Experiments

Loganin, a representative compound in the fruit of Cornus (Cornus officinalis), was observed in cell and animal experiments to suppress the process by which fat cells are formed. A joint research team from the Department of Medical Genetics at Ajou University School of Medicine and NINEB, a subsidiary of Ajou University's technology holding company, reported the results in the International Journal of Molecular Sciences.

Expression of 5 Differentiation-Related Genes Reduced

The researchers treated the mouse preadipocyte cell line 3T3-L1 and primary-cultured adipose-derived stem cells (ADSCs) with loganin while differentiating them into fat cells. The treated cells accumulated fewer lipid droplets, and expression of PPARγ and C/EBPα, which direct differentiation, the fat-synthesis enzyme FASN, the fat-storage protein PLIN2, and the fat-synthesis regulator SREBP1 was also lowered together.

Two obese mouse models were used in the animal experiments: ovariectomised mice (OVX) and mice fed a high-fat diet (HFD). Mice given loganin orally gained less weight, and tissue examination showed less fatty liver and less enlargement of fat cells. The ovariectomy model is used to reproduce metabolic changes after menopause.

From Quality Marker to Research Subject

Loganin is an iridoid glycoside-class compound that has been used as a marker compound to distinguish Cornus from other fruits and to gauge its quality. The significance of this study is that this one compound alone was seen to act on the fat cell differentiation pathway. The doses used in mice cannot be translated directly to human intake, so evidence in people must be weighed separately.

Key points
  • Reduced lipid droplet accumulation and expression of 5 differentiation genes in 3T3-L1 cells and adipose-derived stem cells
  • Suppressed weight gain and fatty liver in two obese mouse models, ovariectomised and high-fat diet
  • Joint research by the Department of Medical Genetics at Ajou University School of Medicine and NINEB

Limitations These are cell and mouse results; effects in people and appropriate intake were not addressed.
